Can 4.3 skins work with 4.2 store?

I know with very extensive customizations they can work. I am looking if they can work with simple, few lines of customizations since both are supposed to be css based anyway?

It would depend on the skin, but I would say generally no.
Sure, both 4.2 and 4.3 are css based, tableless templates - there are changes/improvements in 4.3 over 4.2.
E.g in 4.3, the head template pulls in more templates, whereas it doesn't in 4.2.
So the structure of 4.3 templates are different to 4.2.
